キャンペーンは、基本的に安全地帯が無く自分のフットワークやエイムの精度がものをいう。
硬い敵やタレットを用いたシューティングなどは、Lトリガー連打でエイムアシストを連発すると楽になる。

・キャンペーン実績関連
上位難易度クリアーで下位難易度実績も一斉に解除される。
インセインは、難易度不問で1周クリアーすることでアンロックされる。
また、カウント系や収集系の実績は、チェックポイント通過などのセーブされるタイミングで記憶される。
そのため、足りない部分は任意のチャプターから再開することで補完が可能。
・Hippy-Stomper
ネズミなど元から小さい敵、Shrink Rayを当てて小さくなった敵を踏み潰す。
敵が視野に入っていない(真下を向いていない)と踏み潰し攻撃が発動しない。
・Judge, Jury, Executioner
敵が膝崩れの状態、もしくはFreeze Rayで凍らせた状態の時に、
敵に近づいてXボタンでとどめをさす。後者が確実
・Noms
食べ物の種類は問わない、10回食べれば解除。
序盤のデュークタワー内やストリップクラブなどにある自販機前で食いまくる。
・On the Noggin
ピストルは一撃、レールガンによるHSは頭が大きくなったのち破裂する。
・FBT
自分が10回転倒させられる。
アサルトコマンダーの体当たり、エイリアンクイーンの触手攻撃を食らって稼ぐ。
・Turd Burglar
Duke Lives開始直後のトイレの個室に大便が流されず残っているので、それを拾う。
・I Need a Date
ダムに入りFreeze Rayを手に入れて、指紋認証で入るエリアの真正面に位置する事務所内。
壁掛けのカレンダーを全てめくる。
・Balls of Steel
一見難しい条件だが、時間をかければ簡単に100万点に届く抜け道がある。

左上のフリッパーを上げたまま玉をキープし、すぐ左のパネルを倒し続ける。
パネルは1つ300点、3つ揃えて5300点。倍率を上げておけば5分程度で終わる。
・Drawrings
冒頭のスタジアム控え室にあるサイクロイドの描かれた白板に落書きをする。
・Substance Abuser
ステロイド(Dパッド左の強化剤)を服用した後にビール(Dパッド下)を使用する。

デュークのEGOを最大にする。チャプターセレクトから補完可能
EGOを獲得していない状態からスタートする「ニューゲーム」を選択し、上書きセーブしてしまわないよう注意
・Gunslinger
The Duke Caveの道中で手に入る黄金のピストルを、キャンペーンが終了するまで捨てずに持ち込む。
厳密には、各チャプターごとに一貫して黄金銃を持ち歩いていればよいので、
チャプターセレクト→任意のチャプターを開始でも解除可能(初期装備が黄金銃であるため条件を満たせる)。
・Special Thanks
キャンペーンクリアー後のスタッフロールをスキップせずプレスカンファレンスシーンまで待つ。
