NaCl 10g, East extract 5g, Bactotoripton 10g, for plates, additionally, 15
g Bacto Agar
Water (Deionized) 1 littler, also add 5 N NaOH 200 ul (determine the amount
for each batch of East extract. to pH 7), Use magnet/stirrer to dissolve
them.
Autoclave
Allow the solution to cool to a temperatere where you can manage to hold
the bottle. (or about 60deg)
Add appropriate antibiotics (e.g., ampicillin at 50or 100 ug/ml).
Pour about 20 ml per 10cm petridish. (i.e., 1 litter gives 50 plates). You
do not have to actually measure the volume. Simply pour such an amount that
spreads on the entire bottom, which is about 20 ml, although depends on the
temperature (i.e., viscosity of the solution).
Let them cool to r.t. and store in the cold room (4 deg).
